Allele Analysis · CVENTBIO

Discover & Compare Alleles

Analyze retained reads from a completed PCR order. Define your target regions, discover supported variants, or search for expected allele sequences.

Experimental analysis. Detection limits and biological accuracy remain unvalidated. Results depend on read quality and coverage.

From strands to alleles

Three amplicon variants share the same flanks, with TAC, a deletion, or TTC in the target region
1. Mixed ampliconsShared flanks; different targets.
An individual DNA strand passes through a nanopore
2. Sequence strandsRead individual DNA strands.
Separate sequence reads preserve the individual target variants
3. Compare readsGroup supported allele sequences.
Example allele read percentages: A1 70 percent, A2 20 percent, A3 8 percent
4. Reveal allelesSee sequences and read percentages.

Conceptual illustrations; example sequences and percentages.

Understand mixed amplicons

Resolve mixed sequences

See distinct, supported alleles when a mixed Sanger trace is difficult to interpret.

Find sequence changes

Compare substitutions, insertions and deletions against your reference.

Analyze GC-rich targets

GC-rich amplicons can be analyzed with sufficient amplification and usable read coverage.

What your report shows

See each supported allele’s sequence, length, reference differences, supporting reads and read percentage.

Example alignment: A1 ACGTACGTA, A2 ACG---GTA with a three-base deletion, and A3 ACGTTCGTA with a substitution
Compare each allele with the reference. Dashes show deleted bases.

A2237 bp200 / 1,00020%
A3240 bp80 / 1,0008%

2% of usable reads are unassigned in this example. Percentages describe usable reads.

Choose your analysis

Discover alleles

Identify supported sequence variants between your chosen flanks and compare them with a reference.

Find specified alleles

Search for up to four expected allele sequences. Results show detected, not detected or inconclusive when evidence is insufficient.

Get started

  1. Select your order and samples. Use completed full-length PCR sequencing results with retained reads.
  2. Define your target. Enter shared left and right flanks (15–200 bases each) and a reference target (1–1,000 bases).
  3. Run and review. Choose an analysis mode, then view your allele report and download the data.
